Canon
In the film, it is established that Alex and Lara were one-time lovers. Their relationship ended when he stole a pair of prayer wheels from her. Though Alex wants to rekindle the relationship, Lara does not and rebuffs his advances. Their paths cross, however, when she pursues the Triangle of Light (a relic which controls time) and the secret society known as the Illuminati. Alex is employed by Manfred Powell, a member of the Illuminati out to piece together the Triangle for his own personal gain (presumably world domination).
Alex eventually agrees to help Lara defeat Powell and therefore, save the world as we know it. They venture to Siberia where the last piece of the Triangle is hidden somewhere in a lost city. However, in order to coerce Lara to reveal the location of the last piece of the Triangle, Powell stabs Alex. He bleeds to death, as Lara kisses him goodbye, suggesting she still retains feelings for him.
After assembling the Triangle, however, Lara is able to control time, and one of the things that she does is to reverse the knife so it stabs Powell and not Alex. Alex's last canon appearance is trying to convince Lara to leave the collapsing ruins with him, but when he fails to do so, disappearing with the surviving members of their expedition.
Post-Canon
Since time was altered to resurrect him, Alex's death doesn't exist in history. However, he remembers it, and it manifests itself in memories that begin to surface just three days after he leaves Siberia. Convinced these are nightmares and he is losing his mind, he takes a voluntary retirement from tomb raiding to come to grips with his sanity. During this time, he sinks into depression and many of his friends desert him. The only friend to still support him is known 'fixer' Charlie Price, whom Alex leans on more than he'd like to.
However, when he meets Lara Croft again, he knows that she is the only person who can tell him what really happened to him. He agrees to accompany her on a South American journey, believing that he can then get her to reveal the truth about Siberia. When they are recovering in Peru, Lara tells him that what he is experiencing are, in fact, memories and that she did save his life. Confronted with a second chance, Alex begins to mull over what to do with his life -- while rekindling a relationship with Croft.
Background
Alex was born and raised in Boston, Massachusetts. His father was a police detective, who was forced into early retirement due to health problems, and never quite recovered from having lost his job and sense of purpose. Alex's father took over a local bar and became a bitter, cynical man with an adversarial relationship with his son. As soon as he was able, Alex fled Boston. It is not known if his parents are still alive.
However, Alex has taken on a lot of his father's qualities, including his arrogance and his insecurities.
He graduated from Oxford College.
Alex drives a Mercedes and maintains residences in New York and London, though he seems to favor the one in London.
