CCAP S/W

From Protists

  1. Put a layer approximately 1 cm deep of loamy soil (soil in media cabinet) into a 1L media bottle labelled "CCAP S/W".
  2. Add distilled water to a depth of ~10 cm and cover with foil.
  3. Autoclave for 15 minutes at 15psi on each of 2 consecutive days.
  4. Allow to stand a further day before inoculating. pH should be between 7.0 and 8.0. Adjust with 1M HCl or 1M NaOH to appropriate pH if necessary before inoculation. (Probably slightly acidic)
  5. Allow the media to settle before use, using only the supernatant to refresh cultures.
  6. Store in Media cabinet.
