- Put a layer approximately 1 cm deep of loamy soil (soil in media cabinet) into a 1L media bottle labelled "CCAP S/W".
- Add distilled water to a depth of ~10 cm and cover with foil.
- Autoclave for 15 minutes at 15psi on each of 2 consecutive days.
- Allow to stand a further day before inoculating. pH should be between 7.0 and 8.0. Adjust with 1M HCl or 1M NaOH to appropriate pH if necessary before inoculation. (Probably slightly acidic)
- Allow the media to settle before use, using only the supernatant to refresh cultures.
- Store in Media cabinet.
