ATCC 802

1. Make the media solution as follows:

Cerophyll / Cereal Leaves 2.5 g Distilled H2O 1.0 L

a. Add the cerophyll to the distilled water in a flask. b. Add a stir bar and cover the opening of the flask with foil. c. Put the flask on the hot plate. Turn the “Stir” knob up to 6 and the “Heat” knob up to 10. Boil for 5 minutes (will be small bubbles coming up) . d. Add 20 mL of distilled H2O to compensate for evaporation. e. Filter through Whatman #1 filter paper using the ceramic funnel in the media cabinet. f. Add 0.94 g of Na2HPO4.7H2O to the solution. g. Autoclave for 15 minutes at 121°C. h. Label as “ATCC 802” and store in Media cabinet. 2. 24 hours prior to inoculation, add the appropriate bacteria (indicated on “Tina’s Organism List”) for the organism to the media in the tissue flask for culturing. 3. Incubate the tissue flask in the 33.5°C incubator (middle shelf) overnight. The media is now ready to be inoculated with the protist.
