  1. Make the media solution as follows:
    • Cerophyll / Cereal Leaves 2.5 g
    • Distilled H2O 1.0 L
      • Add the cerophyll to the distilled water in a 2L-flask or beaker.
      • Add a stir bar and cover the opening of the flask with foil.
      • Boil for 5 minutes on the hot plate (“Stir” knob at 6; “Heat” knob at 10).
      • Add 20 mL of distilled H2O to compensate for evaporation.
      • Filter through Whatman #1 filter paper.
      • Add 0.94 g of Na2HPO4.7H2O to the solution.
      • Distribute the media into media bottles for autoclaving.
      • Autoclave for 15 minutes at 121°C.
      • Label as “ATCC 802” and store in Media cabinet.
  2. 24 hours prior to inoculation, add the appropriate bacteria (indicated on “Colorless Organisms” list) for the organism to the media in the tissue flask for culturing.
  3. Incubate the tissue flask in the 33.5°C incubator (middle shelf) overnight. The media is now ready to be inoculated with the protist.
